Mixed Culture Lipid Determination

This is a hard one.

Suppose i´ve got two mixed culture samples of algae.

How does one measure the difference in lipid concentration between the two cultures?

Extraction is pointless as you extract everything that is hydrofobic and uncharged. As its mixed culture a straigh comparisson between samples does not work as the composition of the cultures are different. I Dont know what is the follow up step after extraction, the lipids should be purified and then transesterfied. That is measureble. However i dont know much about the purification and transesterfication.

Any idea's on this topic would be great!
