Photo Bio-Reactors

I have been reading over many threads here since my recent joining of this group about PBRs.


The majority of my experience is in cell culture, small scale process development area.


Many years ago I worked with large perfusion type reactors and fed-batch systems.


I  currently work with Bioreactors that range from 50ml up through 8L with high prescision control platforms (process control computers) using the well know PID type control. The company I work with has perfected the ability to regulate light intensity ranges across the color spectrum and they can be dialed in to fit a particular strains need. We also have the ability to cycle "on-off" time and mimick daylight by intensity reduction just as our natural light cycle presents to us on a daily basis..


The light is contained within the PBR and the reactor is then autoclaved with all sensors in place for a sterile environment. Each reactor is then inoculated with a particular strain for contamination free cultures having pH, DO, temp, gassing, agitation and substrate feeding control.


I am currently working with  2 strains at the moment and seeing very good growth rates thus far. I also have the ability to incorporate scripts (VB) into the control logic for precise process development understanding. While this system is not cheap by any means, it provides you the Scientist, the ability to define your process. Knowing what is happening within the culture is the very essence of scale up for mass production.

Dunaliella tertiolecta and Tetraselmis

Dunaliella tertiolecta and Tetraselmis cultivation. Would any of you be able to clarify the sparging of CO2 (pure) into a small scale system? What about the normal CO2 content of air? Although it is rather minimal, would it be enough to sustain say 1L of culture?


I have seen 02 production of 1 ml/min at densities of 1.3 E6/ml. I have also observed N2 being used at a ratio of 24:1 (N2/CO2) to maintain a stable pH of 8.0. Anyone know the N2 or N03 effect on these cell lines?
